Truncation Peptide Library

The purpose of building truncation peptide libraries is to determine the shortest amino acid sequence required for maintaing activity in a peptide. Experts systematically truncate the peptide sequence, that is, remove the flanking residues of the original peptide. To construct this unique library. Generally, an Alanine Scanning is employed to determine the essential amino acids in the peptide, and the truncation direction can then be determined and adjusted to maintain these residues while truncating the opposite end.

Library Design

BOC Sciences employs advanced incremental truncation technology to generate peptide libraries when each onebase-pair in a gene or gene fragment is missing, or detailed structural information is unavailable. Our experts have developed several useful strategies:

  • Combination of α-phosphothioate dNTPs by PCR
  • Time-dependent nuclease method
  • Primer extensions (THIO(pcr) truncation and THIO(extension) truncation, respectively)
