Aiming to accurately identify residues that play a role in peptide activity, scientific experts have employed many strategies to design and synthesize alanine scanning libraries. Alanine can be used to replace non-alanine residues of the original peptide. The measurement of the corresponding change in epitope activity can reflect the contribution of each residue to the function of a given protein, helping to identify the specific amino acid residues that are closely related to peptide function, stability and conformation.

BOC Sciences has introduced this powerful tool to construct our unique libraries to identify B-cell epitopes.
Firstly, each non-alanine residue of the peptide will be substituted sequentially by one alanine at a time by employing a a highly specified site-directed mutagenesis approach
Then, a large number of plasmid clones will be expressed accurately and rapidly
Finally, we measure the corresponding changes in peptide-antibody binding activity
